Tunis to Douz is about 426 km in a straight line and around six hours by road — so the Sahara is not a day trip from Tunis, whatever a listing may imply. Two days is the realistic minimum, three if you want the Star Wars sets or the mountain oases as well. Tozeur, the other desert gateway, is about 371 km.

It depends what you mean by “the Sahara”. The dunes proper — the Grand Erg Oriental — begin at Douz. The oasis towns of Tozeur and Nefta, the mountain oases and the salt lake sit a little north of them, and most visitors call all of it the desert. Measured from the Tunis medina in a straight line (road distances are longer, and they are the ones that fill the day):

Roughly six hours from Tunis to Douz without stops, and nobody drives it without stops. Two routes are used. The inland route runs through Kairouan, Sbeitla and Gafsa to Tozeur; the coastal route follows the motorway south past El Jem and Sfax, then turns inland at Gabès for Matmata and Douz. The Chott el Jerid causeway lies between Tozeur and Douz, so any circuit that visits both crosses it — the one stretch of the journey worth slowing down for.
The practical consequence is that a two-day trip from Tunis spends most of its first day and most of its last in the car. That is fine if you plan for it, and a disappointment if a listing promised “two days in the Sahara” and meant one evening of it. The stops that make the drive worth having — Kairouan, El Jem, the Chott — are what the longer circuits are built around.

Domestic flights to Tozeur–Nefta airport exist, though the timetable is thin and it is worth checking current schedules before building a trip on one. Long-distance buses and louages (shared taxis) reach Tozeur and Douz, but the sites themselves — the mountain oases, the film sets, the camps — are off any scheduled route: from the town you are hiring a local driver, usually with a 4x4. Renting a car works for the paved roads, but fuel stations thin out in the south and the last approaches to the camps are piste.
A private car with a driver is the only option that covers the whole route in one booking, and it lets the circuit stop at El Jem or Kairouan instead of treating the drive as dead time. More on the options in Tunisia without renting a car.
October to April is the season. Nights in the desert are cold from November to February — a camp night in December is beautiful and needs real layers. From May to September the far south regularly passes 40°C at midday and reaches 45°C, and the days are best spent indoors. Not sensibly. The round trip is over 800 km of driving, which leaves almost no time in the desert. Every honest itinerary from Tunis is two days or more.

It can be, if you only want the oases and have little time. But there are no transfer fares published for Tozeur airport here, and a private circuit by road lets you stop at El Jem, Kairouan and the Chott on the way.
About the same as to Douz: plan a full day of travel, with a stop at Kairouan or El Jem to break it up. Tozeur is the nearer of the two desert gateways in a straight line, but the road is not faster.
October, November, March and April are the most comfortable: warm days, cool nights. December to February is beautiful and cold after dark. From May to September it is very hot in the south.
